LC3 - Hallmark Target of Autophagy

 
A cell in distress can respond in several fashions, one being the autophagy pathway. Such reactions can be induced or upregulated by external stress factors such as starvation, hormonal imbalance, oxidation, extreme temperature and infection.

Autophagy can be regarded as a catabolic trafficking pathway for bulk destruction/turnover of long-lived proteins and organelles, carried out via lysosomal degradation.
 
The key event is the formation of the cytoplasmic autophagosome, which engulfs proteins destined for destruction, eventually fusing with the lysosome for completion of the degradation process.

This transient autophagosome is always detected by its heavy association with the protein LC3, which thus can be used as a reliable indicator of autophagic activity.

LC3 Western Blot Tip:

 
LC3-I is converted to LC3-II via a phosphatidylethanolamine (PE) modification during autophagy induction. LC3-II may not adhere well to a nitrocellulose membrane due to the enhanced hydrophobicity of the PE modification. Always use a PVDF membrane to assure optimal results!
